Description: Cilnidipine is a 4" generation Calcium channel blocker. It is vaso selective and has a weak direct dromotropic effect, a strong vasodepressor effect and an arrhythmia inhibiting effect
Mode of action: Cinidipine is a Cal channel blocker with a dual L & N-type Ca channel-blocking action The L-type calcium channel blockade by ciinidipine affects predominantly vascular smooth muscle, thereby producing vasodilation of peripheral resistance vessels and coronary arteries. The blockade of N-type Ca channels effectively suppresses neurohormonal regulation in the cardiovascular system, including sympathetic nervous system and renin-angiotensin-aldosterone system. Thus, cinidipine can reduce various types of complications of hypertension as well as cardiovascular diseases.
Pharmacokinetics: Cinidipine has a good oral absorption and a long time of action. After oral
administration, drug concentrations reaches peak level at 1.8 to 2.2 h and show a long half-life of 7.5 h
Cinidipine was rapidly metabolized in human liver microsomes and dehydrogenation of dihydropyridine
ring of cinidipine is crucial for the elimination of cinidipine. Cytochrome P450 3A(CYP3A) is the major
human CYP involved in the dehydrogenation of dihydropyridine ring of citnidipino.
Composition: Cildip 5 mg Tablet: Each film-coated tablet contains Cinidipine INN 5 mg. Cildip 10 mg Tablet: Each film-coated tablet contains Cinidipine INN 10 mg. Dosage & administration: The recommended adult oral dosage of Clinidipine is 5-10 mg once daily.
Indications: Hypertension.
The dosage can be increased up to 20 mg in a day, if needed. Contraindications: Cardiogenic shock recent MI or acute unstable angina, severe aortic stenosis.
Side effects: Dizziness, flushing, headache, hypotension, palpitations, Gl disturbances, myalgia, tremor
Use in pregnancy & lactation: No specific information about US FDA pregnancy category. Caution should be exercised during Cinidipine use in pregnancy Nursing mothers should consult a physician before taking Cilnidipine.
Precautions: Hypotension, poor cardiac reserve, heart failure. Sudden withdrawal may exacerbate
angina. Discontinue in patients who experience ischemic pain following administration Drug Interactions: It interacts with other antihypertensive, aldesleukin, antipsychotics that cause
hypotension, may modify insulin and glucose responses, quinidine, carbamazepine, phenytoin,
rifampicin, cimetidine, erythromycin.
Over dosage: Available data suggest that gross over dosage could result in excessive peripheral vasodilatation and possibly reflex tachycardia.
Storage: Keep out of reach of children. Store in a dry place, below 25°C temperature and protected from light.
